Deep sequencing of the mantle transcriptome of the great scallop Pecten maximus ArchiMer

RNA-Seq transcriptome data were generated from mantle tissue of the great scallop, Pecten maximus. The consensus data were produced from a time course series of animals subjected to a 56-day thermal challenge at 3 different temperatures. A total of 26,064 contigs were assembled de novo, providing a useful resource for both the aquaculture community and researchers with an interest in mollusc shell production.

In every organism, evolution and acclimation to environmental changes are orchestrated by numerous molecular players. Among them, transcription factors (TFs) play a crucial role by regulating gene expression. Therefore, identify TFs involved in the production of high value products is a significant step in a biotechnological context. The laboratory has at its disposal a mutant strain of the haptophyte microalga Tisochrysis lutea producing twice more storage lipids than the wild type strain when exposed to nitrogen deprivation. Highly unsaturated fatty acids of the omega-3 series (HUFA) are major constituents of cell membranes, yet poorly synthesised de novo by consumers. Their production, mainly supported by aquatic microalgae, has been decreasing with global change. Understanding the consequences of such reductions is essential for ectotherm consumers, since temperature tightly regulates the HUFA content in cell membranes, maintaining their functionality. Crassostrea gigas is the main species of oyster cultivated in the world. Since 2008, mass mortality events have been affecting oysters aged less than one year old in Europe and Oceania and have been associated with the emergence of the Ostreid herpes virus μVar (OsHV-1 μVar). In Europe, these events are seasonal and occur when the seawater temperature is between 16°C and 24°C. In this work, the effect of high temperatures (21°C, 26°C and 29°C) was evaluated on the susceptibility of oysters to OsHV- 1 but also on the virulence of virus.High temperatures (29°C) reduce the susceptibility of oysters to OsHV-1 without altering the infectivity of the virus and its virulence. O presente estudo teve como objetivo comparar o transcriptoma abomasal de ovinos da raça Morada Nova com fenótipos divergentes de resistência a Haemonchus contortus. Para tanto, 287 cordeiros foram submetidos a dois desafios parasitários experimentais e monitorados quanto ao ganho de peso, ao volume globular e à contagem de ovos por grama de fezes. Após análise de dados, foram selecionados 10 animais de cada extremo de fenótipo de resistência parasitária, sendo que metade desses animais foi submetida ao sequenciamento de RNA (RNAseq) da mucosa abomasal. In 2005, draft sequences of the genomes of Trypanosoma brucei, Trypanosoma cruzi and Leishmania major, also known as the Tri-Tryp genomes, were published. These protozoan parasites are the causative agents of three distinct insect-borne diseases, namely sleeping sickness, Chagas disease and leishmaniasis, all with a worldwide distribution. Despite the large estimated evolutionary distance among them, a conserved core of ~6,200 trypanosomatid genes was found among the Tri-Tryp genomes. Extensive analysis of these genomic sequences has greatly increased our understanding of the biology of these parasites and their host-parasite interactions.
